I recall an incident while at a summer program a few years ago. We, the students, were staying overnight on a college campus to participate in seminars that would further our growth and readiness for the future. As part of the program, we were required to stay overnight in the dorm rooms. We were each paired up with one other person who would be our roommate for the two days. I come from a neighborhood that is primarily white so I was pleasantly surprised to learn that I would be sharing a room with someone who was from the Middle East. We quickly settled into our rooms and prepared ourselves for the first of two days of learning activities. My roommate awoke, got up and spread a blanket on the floor to pray. I watched my roommate kneeling on the floor saying words in a language that I didn’t understand. When finished, I asked my roommate about this practice and learned a little bit about the Muslim religion and why prayers are said several times daily. Many older couples who had delayed having children began having them when the war ended. This increase in births among both younger and older American couples created the Baby Boom Generation. This large group of people born in the U.S. from 1946 to 1964 amounted to 76 million children. Those born during this time make up approximately 36% of today’s population (Deutsch). What impact has this had on the economy? Read on to see how the baby boom generation has affected the economy and what will happen as they retire. This generation has had a huge impact on the economy due to its sheer size. At first there was a crunch at hospital maternity wards. When the boomers reached school age, schools became overcrowded, forcing towns to build additions. By the time they were entering senior high the cities and towns were expanding. New office buildings, hotels, apartment complexes and stores were springing up everywhere. When they reached college age, American Higher Education had to expand. Large enrollments along with inflation pushed college costs higher. When they started families of their own and became homeowners it drove real estate prices up. We can expect a large demand for retirement housing, elder care and finally the funeral industry as the Boomers continue their life cycle. Marketers have followed the boom generation for decades, producing items that appealed to them as children, teens and young adults. With more money in their pockets than time on their hands, today’s baby boomers are dining out at an all time high. Now it is people over the age of 40 who have the resources to purchase big-ticket items. Today the engine that is driving our economy and our stock market is due to the lopsided demographics present right now. â€Å"To His Coy Mistress† is primarily the author, Andrew Marvell, trying to convince and seduce â€Å"his coy mistress†, into having intimate relations with him. The poem has three stanzas; each with a different purpose: the first stanza gently and subtly flatters his mistress, using positive diction and images to show, how Marvell wishes he could love her for all of eternity; the second stanza, however, uses imagery to show how time is moving fast and also, strongly negative diction and images to show how life must be lived happily, for there is no chance to after death; the last stanza, the conclusion of the poem, uses quite sexual images to tell his mistress, that because time is limited, they should make the most of it, and enjoy life’s intimate pleasures together. Imagery and diction have been used effectively throughout the poem, to achieve the author’s purpose, of seducing this lady. The author also conveys a theme throughout the poem; life is sho rt, your time on earth is limited, and therefore we must make the most of life’s pleasures while we still can. In the first stanza, imagery and diction, flatters this lady, Andrew Marvell wishes to seduce, and depicts his great and ever-growing love for her. Marvell begins by describing how ideally he would have â€Å"world enough and time† to love this lady. They would â€Å"sit down, and think which way to walk and pass [their] long love’s day.† Imagery shows them taking their love very slowly. This image is created by diction with relaxed and slow connotations, such as â€Å"sit down†, and â€Å"walk.† The diction within that line, also creates alliteration, â€Å"which way to walk†, and also, â€Å"long love’s day;† this alliteration, and the long vowel sounds in â€Å"way†, â€Å"walk†, and â€Å"long†, creates a slow and steady rhythm, and a relaxed mood and tone to the stanza, which allows the author to convey to â€Å"his mistress† that he wishes they could take their love slowly and steadily. Later on in the stanza, he uses diction to create images, to flatter â€Å"his mistress.† â€Å"Thou by the Indian Ganges’ side shouldst rubies find; I by the tide of the Humber would complain.† The imagery shows how he sees his mistress as exotic, by comparing her to the Indian Ganges’, which at that time, was an faraway and exotic place; while, comparing himself with â€Å"the  Humber†, he views himself as ordinary, compared to her. The effect is that it fulfils the author’s purpose for it, which was to flatter this lady. The diction also helps him achieve this, â€Å"shouldst rubies find†; rubies are precious and beautiful, and by using this diction, he again flatters her, by describing how beautiful and precious she is to him. In the first stanza the author has used diction and imagery effectively to create a relaxed and easy mood and tone, to show how the author wishes he can just slowly and eternally love this woman; a â€Å"state† which she â€Å"deserves.† He also achieves his purpose of gracefully complementing this lady on her beauty, in more ways than physically. In the next stanza, Marvell uses diction and imagery to show how there is nothing to be enjoyed in the eternity of death, and how death is a lonely place, therefore another person’s love must be experienced during life. He tells us that â€Å"at [his] back [he] always hears Time’s wingà ¨d chariot hurrying near.† â€Å"Winged†: this diction gives us an impression that the â€Å"chariot† is quick, and therefore the imagery, created by the personification of â€Å"Time†, shows that time travels quickly; life is short. â€Å"Yonder before us lie deserts of vast eternity.† I believe the â€Å"deserts of vast eternity† metaphorically symbolises death. â€Å"Deserts† suggests lifeless, desolate; while â€Å"vast eternity† uses long vowels sounds in â€Å"vast†, combined with the ‘e’ sound being repeated and carried on at the end, in â€Å"eternity†. The combined effect of the diction: an image, showing the boring, lifelessness of death. This is summed up at the end of the stanza: â€Å"the grave’s a fine and private place, but none, I think, do there embrace:† love and its pleasures may only be experience during life. He also tries to convince â€Å"his mistress†, that keeping her virginity, is a silly thing to do. Marvell refers to â€Å"long preserved virginity† as a â€Å"quaint honour†; the choice of diction, by using quaint, shows the author’s negative tone towards keeping your â€Å"virginity†: it is too old-fashioned, odd, and somewhat of a joke. The diction and imagery in this paragraph show us that life must be enjoyed, for such pleasures do not exist after death. In the last paragraph, Marvell, uses sexual, passionate diction and imagery  to show that to enjoy life to the fullest, they must have intimate relations together. â€Å"Now therefore, while the youthful hue sit on the skin like morning dew, and while thy willing soul transpires at every pore with instant fires†; the simile creates an image, showing us that this lady is physically young, and the metaphor shows us that she is either blushing, or seems to be very excited, or both. Marvell uses this imagery to subtly tell this young woman, that it is obvious, she wants the same as him. â€Å"Now let us sport us while we may, and now like amorous birds of prey†; the diction, amorous, has very passionate connotations, even more so than love, and the simile, like birds of prey, suggests a physical side of love, and creates an image of a fearless bird, diving as soon as it sees a chance for kill; therefore the image shows us, that while we are able to, we must make the most of the physical and passionate pleasures of life, without too much concern, and also most importantly, as soon as we may. The tone and mood, created by the diction and images, in the paragraph is very positive, and passionate, and its purpose is to convince â€Å"his mistress† that what he is suggesting, is the right thing to do, and to do so without any worries, and as soon as possible. We must enjoy the pleasures of life, while we may, for that is impossible after death. Andrew Marvell effectively uses diction and imagery throughout this poem, to convince and seduce a young lady, into having physical relations with him. He uses diction and imagery in the first paragraph, showing how, greatly he loves her, and how willing he is to only love in a non-physical way, till the â€Å"last age† had they all of eternity. However, in the second paragraph, negative diction and images, show us that time is limited, and the pleasures of life, cannot be found in after death; therefore we must enjoy them while we live. The third and final paragraph, is the conclusion to Marvell’s argument; he uses passion filled images and diction, to show that they should therefore engage in a physical and intimate relationship, for this maybe the only opportunity they ever get. A theme conveyed is that we must enjoy all of life’s pleasures, for we only get one chance to live.

Chapter VIII is about signal units involved during World War II. Throughout each phase of the conflict, new problems and difficulties arose that could only be overcome through hard work, ingenuity and extreme bravery. The huge scope of the war on numerous continents tested the will and determination of the signal corps. 2. The first real need for signal support was hemisphere defense. This included protecting the Panama Canal from bombings, the British supply line by way of Greenland and the Japanese invasion of parts of Alaska after bombing of Pearl Harbor. They installed radar along the coasts and ran thousands of miles of wire through the frozen terrain of Alaska. On Puerto Rico, Signal Units provided aircraft warnings and communication support to help protect the Caribbean. 3. The first Signal units to actually face the enemy were in the Pacific. After the Pearl Harbor bombing, the Japanese began invading the Philippine Islands. They forced General McArthur to retreat to the Bataan Peninsula and finally Corregidor. Not all of the soldiers could get away. Lt. Gen. Wainwright was in command of the garrison and ventually moved his headquarters underground into the Malinta Tunnel. Col. Theodore Teague was commanding signal officer and once the Japanese invaded the tunnels, he ordered his signal soldiers to destroy all equipment. To keep communications going in the Philippines, the Signal Corps created the 978th Signal Service Company out of largely Filipino volunteers. They worked hand in hand with the guerilla fghters that resisted the Japanese invaders. These volunteers helped pave the way for the eventual Allied liberation of the Philippines. 4. The Signal Corps became involved in a grander scale of combat operations in North Africa. General Dwight D Eisenhower was the overall Allied Commander of invasions of Morocco, Algeria and Tunisia. His chief Signal Officer was Brig. Gen. Jerry V MateJka. The first big test was operation TORCH, in which the Signal Corp was to provide communications for the amphibious assaults. Numerous problems arose from failure to keep the sea water from damaging equipment to the equipment itself being too heavy. They eventually set up permanent ACAN stations. Another issue was the different codes and call signs used by the British Army and Navvy, Royal Air Force and the Army Air Forces. Signal soldiers had to be adaptive and learn on their feet. 5. In helping to plan the Invasion of Normandy on the European front, the Signal Corps became part ot the Services ot Supply. Brig. Gen. illi m S Rumbougn became the theaters chief signal officer. He was faced with signal planning for the largest military operation of the war. This included establishing system of priorities for telephone traffic and allocating frequencies for the 90,000 transmitters needed for the operation. He also created the Joint Assault Signal Company OASCO) to provide signal support for the amphibious assaults at Omaha and Utah beaches. Also, wenty-eight signal men from the 101st Airborne Signal Company were the first signal men to land in France. They flew in with their SCR-449 radios on gliders. Multichannel radios were first incorporated in the Normandy invasion. 6. Once Hitler committed suicide and Germany surrendered, the Signal Corps put all their resources in support of the Pacific campaign to finish off the Japanese. The jungles of the numerous islands provided many issues with radio communications. To get the message through, miles and miles of wire were run through the trees without proper equipment. The SCR 300 radio was used to communicate with aircraft or arrangement of supply drops for the advancing Allied forces. The Signal Corps had also built their own signal fleet equipped with VHF radio relay sets. Their flagship, the Apache, was specifically devoted to public relations. After the ferocious battle of Iwo Jima, the Signal Corps began planning the communications for the proposed invasion of Japan called Operation OLYMPIC. The dropping of atomic bombs on Hiroshima and Nagasaki brought the war to a quick conclusion. The Signal Corps performed their last duty by broadcasting the formal Japanese surrender proceedings aboard the USS Missouri to all the world.

Since these allegations arose back in 2010, he has lost many followers and offended many other cancer survivors, with what the world looks at as cheating. Lance Armstrong Is the spokesperson for his own organization, the Lance Armstrong Foundation (ALFA). The ALFA Is known for their bracelets which over 65 million of these were sold, making his organization very well known and well respected. When Armstrong came out of retirement and returned to professional cycling In 2009, the ALFA revenues increased from $32. 6 million in 2008 to $41. 7 million in 2009 (Maybe Its About the Bike, peg. 2). Also, the Sports Illustrated writer Selene Roberts reported four massive accusations about Armstrong, all of which are plenty enough reason to force the public to look at Armstrong as a disgrace and a cheater. She even referenced the similarities between his and Roger Clemens doping scandals. For a personal perspective, Lance Armstrong has always been considered a hero. He was a man who did not let cancer deny him from getting back on his bike and winning any biking titles. Unfortunately for Mr.. Armstrong, he ruined his image and will probably never be able to regain such a strong relationship with his admirers ever again. There Is no reason why Lance should have allowed himself to compete knowing he was galling an unfair advantage. What Is even more unjust about the situation is the fact that after 60 Minutes aired an interview with his former teammate, Tyler Hamilton. Hamilton, one of Armstrongs former USPS teammates, publicly stated that he had first-hand knowledge that Armstrong had doped in the dead-up to at least three of his Tour De France victories (Maybe Its About the Bike, peg. 2). Its almost disturbing that even after such a valuable source threw Lance under the bus he could still proclaim his innocence. There is no soft spot for anyone who allows themselves to rob hundreds of athletes for what they have also worked day and night for. It Is my belief that Lance Armstrong was rightfully stripped of his titles, and should never be allowed to compete again. Many supporters might argue that he ad cancer and deserved create a more even playing field for himself. This Is not the case, If Lance Armstrong, one of the greatest athletes of all time, wanted to be remembered forever he should have thought about the consequences before he cantata. My Family Case Study will examine the ‘Nguyen’ family and their current struggle with family finances. I will present the family, their history, and their response to the challenge of dealing with the crisis related to the family finances. I will present the family situation when they were in Vietnam first. In Vietnam, they bought the house and they didn’t have to pay any tax or fee. They were considered as a middle class well-living family in Vietnam. They got their own business over there. Their income per month were enough for them for a good living life. Only the father worked and the mother stayed home and took care the kids. One day they received a letter from the United State Citizenship and Immigration Service (USCIS) said that they got approved to go to America. They got stressful since they got the letter. At that time, they were unsure whether or not to go to the U.S.A or stay back in Vietnam. It was a difficult decision for them because if they went to the U.S.A they had to give up their business which they had built up since they got married. Time went by, they determined to leave everything behind and went to the U.S.A for their kids’ future because they believed that America has a bes t educational system in the world. Until they came to America three years ago, their life had a big changed. Mr.Nguyen was a 45 years old man. He is married to Mrs. Nguyen who was a 40 years old. They were married 16 years ago in Vietnam. They had two children. One named Peter age 5 who was in pre-school and the other one named Mike age 9 who was in intermediate school. One month after they came to the U.S.A, they found a job and got pay minimum wage around 8 dollars per hour. The family lived in an apartment which 2 bedroom. ... ...getting a bad day at work, and feel like he couldn’t make more money to support his family well enough. He sometimes wants to buy electronic things, food, candy, new clothes etc†¦ for his wife and his children but he cannot afford it so whenever the fight starting between him and her, he cannot control all the stress and anxious in his mind and fight back at her even though he doesn’t want to. Mrs. Nguyen wants to help her husband coping with all the stress he has, so she gets some guidelines from the family therapist. Mrs. Nguyen want Mr. Nguyen to spend time with his family on the weekend and not going for a part time job. They will spend their time with their children and planning to have a family vacation, doing activities together or having dinner together in order to maintain a good relationship and not letting financial be a stressful problem in their family.

Just Be Nice

James Q. Wilson have described as the elevation of self expression over and over self control. Such as a black child was discipline in a Connecticut (1996)for wearing his pants sagging. Stephen Carter’s summary â€Å"Just Be Nice† starts out by talking about how life was different back in the day compared to how life is today. Children were taught right and wrong, yes mam and no sir and just simple rules of etiquette in school. Now teachers have to be very careful on what they talk about with students. He believes that if these types of things were taught in school more openly today, then everything would be less violent.Carter then goes on to explain the reasons he feels that society is not trying as hard as it could be. He believes people are more focused on trivial things, then the important things. Carter feels schools have gotten rid of greatly needed academic school programs for less academic inclined programs. Carter makes several very good points on why society in this age is vastly different than what it was decades ago. He believes that society as a whole group must work together to handle things better, so that society may learn to love each one another again and not just tolerate other people.Carter also believes people are mean to each other because they have the right to be and can get away with it. The constitution is there for a reason, but sometimes those rights can be used and abused. â€Å"We have let cynicism replace civility. † (672). The world is a bit cynical. Carter explains that even the simplest gesture can become a problem. He feels that people are afraid to do anything nice, for fear of it back firing. Everyone is suing everyone, and that has major consequences. Accidents happen, so sometimes you just have to trust that they didn’t mean to hurt you.There are people in this world that do abuse our freedom of speech, like Carter says. Many children (and adults) cuss severely on a day to day basis, and need t o understand that language is a key element in what happens next in life. If a person talks foul, then they receive foulness. If you talk nice, however, you receive niceness back. It all depends on how you act and react to things. If we as a whole try to improve our foul mouths and attitude’s, the world would be a happier place. Cussing and acting up in public is very inappropriate and people should respect other people by not doing it or trying to dial it down.It is hard to do because this generation grew up doing it and habits are very hard to break. Carter makes some very good points in throughout his essay. Everyone on Earth is raised differently, so no two people act the same. But regardless of where we live, who we are and how we grew up, we are all taught one very specific and golden rule, â€Å"Treat others as you would want to be treated and have respect your elders. † Stephen L. Carter: Just Be Nice In the essay Just Be Nice, written for the Yale Alumni Magaz ine and later included in â€Å"Civility Manners, Morals, and the Etiquette of Democracy†, Stephen L.Carter addresses several issues in today’s society. He describes the differences in common courtesy, freedom of Speech, and polite etiquette, while expressing his opinions on manners between the older generation and the new age generation in today’s society. Carter also compares the normality’s of today’s society to the time when he was growing up. He partly blames society’s etiquette problem on this generations youth and school systems, he suggests that schools are not disciplining children like they used to and are always under constant scrutiny by todays parents.Carter states that when he went to school in the 60’s teachers and parents were different then they are today. He implies that the modern person takes hard earned rights for granted and gives examples that show people today do not understand the difference between a want and a right. Just because you want to do something, does not necessarily mean it is your right to do it. In his article, Carter compares life today to life in the past. At the heart of heart Carter’s argument are the notions of manners, civility and differences in generational ages.Carter believes that the standards of civility have declined immensely; people have forgotten how to conduct themselves and don’t trust anyone. He believes if we â€Å"place our trust in the vague and conversation stifling language of rights instead. )((paragraph. 4). † This means that if we trusted each other more, then everyone would be more civil with each other. The author states that nowadays when a school tries to discipline misbehaving youths or teach simple rules on manners, it is a crime and it makes news. Carter implies that when he was a child, schools would have classes dedicated to manners, and parents trusted the school’s judgment.

Helping The Homeless Essay

Do you know how many people there are in the world with no home or money? There are millions and millions of homeless people in the world, and I want to help put a stop to it. I feel strongly about this because i grew up in Thailand when i was younger and seeing all these homeless people really impacted my life and how i thought about people. The three best ways that people can help the homeless are to help build shelters so that they are safe at night, to donate food so they don’t go hungry and die, and to donate money so that they can afford food and clothes. There are several ways to help the homeless, one is to help the organizations, that help to homeless, is to donate money to them so they can keep helping the homeless. Or actually go with the organization and build the shelter with them. Maybe, in your opinion, this isn’t the best way to help. But I think donating food for the homeless is the most important thing you can do. Donating food to the homeless is impor tant because it makes so that they don’t go hungry and die. It is one of the most important things you can do, because without food they don’t get nutrition and won’t be able to grow, mentally or physically. And if people don’t have enough food, the will starve, and eventually die. When i lived in Thailand, every time my family and I went to McDonalds, Burger King, or any other fast food restaurant we would buy an extra meal for a 12 year old kid that sat at the same light post every day for 2 years. And every time we gave him food, he would be so happy. It just made me feel amazing being able to make someone, living the life that they did, feel so happy. And i know my parents and siblings felt the same way. And i know that anyone else who helps the homeless will also get the same feeling. But you don’t have to only donate in person, there are hundreds of organizations who help the homeless, poor, neglected members of our society. And in my opinion every one has to do what they can to help. Something that Kyle Tyrrel said â€Å"Seeing a homeless person begging out in the cold, steam escaping their mouths as they ask for money, touches something primal within me. They are alive in the same way I am.† What he is saying in thais is that they breath, just like you breath. They need to eat, just like you need to eat. And they are people, just like you. Donating money to homeless people is important because it makes so that they can afford food and cloths. This is important  because some people wear the same cloths and never have money for new ones. When i was younger and I lived in Thailand I put money in donation boxes or gave to homeless people themselves whenever my family and I went to the Mall or to get dinner. I would do this all the time because i felt bad for people, especially children, who did not have the privileges that i had growing up. And i felt bad for people who were born into that life. Yes some of the homeless have to give all their earnings to someone who has pretty much ‘enslaved’ them, but that doesn’t mean you can’t help in other ways. When i did this i learned how good it felt to help people survive. Because even at a young age i learned that every dollar helps. And this is a really important lesson for our generation, some people just don’t understand what it’s like to be â€Å"The other half† of the world. Donating money is not only good for the people you are donating it to, but it helps you become a better person, and teaches you kindness, and makes other respect your more as an individual. Building homes and/or shelters for homeless people is important because it helps to increase their safety. This is really important, not many people realize that over a billion people have no real home at night. One way that i helped increase the safety of a homeless family was When I was 8 years old in Sweden and my momâ €™s friend in Thailand was helping out this family whose dad had run away when their daughter was born with a sickness and we flew over to help that family and build a house for them. It increased their safety and the mother could leave her 14 year old daughter, who couldn’t walk, at home alone while she got medicine and food. Even though there might be other ways to help homeless people survive and be happy, I think that this is one of the best. And while helping the family and building their new home i learned how good it felt to help others and i learned how â€Å"The other half lived† at a young age which influenced how i think today. This is important for people to learn at a young age because it teaches them not to be greedy and to be happy for what they have. There are lots of different ways to help the homeless, here is another way to help with the building of shelters. Join one of the many, many organizations that build homes for the homeless. Habitat for humanity for example. At my school there is a ‘Habitat for Humanity’ Club. Everyone in it is doing their part, Other people should do their part too. There are millions and millions of less privileged people out there with no food, money or home. And the three  best ways to help them are to donate food, Give the money, And to build them shelters. Homelessness is a huge problem in the world. And i think everyone should do their part and help decrease homelessness. You can donate a few dollars, or build your own homeless shelter, anything and everything helps.

Sample Turnaround Times for Three Histoprocessing Methods Introduction Turnaround time for any pathological laboratory is very important which depends upon the preparation and diagnosis of the histopathologic material. The rapidity advantages the clinician to treat acutely ill patients and influence the work practice of the pathologist. With the advent of modernization tissue processing is modified from the point of tissue removal to embedding for instant histopathological diagnosis by various techniques or methods. After the surgical removal, the tissue undergoes preparatory protocol for preparation of sections which usually involves impregnation with a suitable supporting medium. The stages of tissue processing include fixation, dehydration, clearing, impregnation and embedding for designated durations of time to ensure completion of the procedure. Culling 1974, Bancroft Gamble 2002 The reproducibility and relatively low expense attached to the most commonly employed method continues to recommend it as a valuable tool after nearly 100 years of existence. But with the demand of faster or early reporting, newer techniques like rapid manual and microwave processing are getting introduced. Each of them is unique with their own set of advantages and disadvantages. The conventional tissue processing is reliable and cost effective but time consumption, reagent toxicity and delay in providing diagnosis are the major disadvantages. The rapid manual tissue processing has major disadvantages like the use of noxious chemicals, greater degree of tissue distortion and shrinkage which led to exploration for new short processing schedules. The microwave tissue processing eliminates the use of noxious chemicals, causes lesser distortion of tissue and has shorter processing time but the cost involved in instrumentation is very high. (Panja et al. 2007) Microwaves were invented by Percy Spencer in 1945 which work on the principle of producing heat by oscillating or exciting polar molecules. The microwave irradiation forces dipolar molecules of proteins to rotate through 180Â ° at the rate of 2.45 billion cycles per second (Srinivasan, Sedmak Jewell 2002, Bancroft Gamble 2002). These excited molecules due to kinetics cause collision with adjacent molecules resulting in transfer of rotational energy. This friction causes production of heat within the material itself leading to accelerated diffusion of processing fluids hence faster processing is possible. The advantages associated with microwave processing led to the production of commercially available microwaves specifically designed for tissue processing, however, the cost involved in these is very high (Leong 2004, Rohr et al. 2001). Domestic microwaves are readily available, affordable and had been used for tissue processing with good results earlier by some authors. Thus, the aim of the present study was to compare and analyse the efficacy of three histoprocessing methods and to determine the impact on turnaround times of tissue processing by these three methods. Materials and method In the present study, 60 specimens were selected randomly. The soft tissue specimen fixed in 10% NBF for 24h were included in the study and hard tissues like cartilage, bone and tooth were not included in the study. The gross features of the specimen were recorded and tissues were cut into three pieces of approximately same size to be processed by three methods. The sections obtained after processing were subsequently stained with HE by routine and microwave staining method. The stained slides in each group processed by three techniques were randomly numbered for a blind study and circulated among four observers referred as O1-O4. The observers graded each parameter on the format given in appendix D by following specific criteria as given in appendix C. Methodology Microwave tissue processing The microwave oven was calibrated as the microwave energy is non uniform within the chamber. Thus, hot and cold spots were detected in the chamber with the use of thermal paper sheet instead of the use of extra water load as suggested by various authors. The cold spot provided the most consistent results every time. The technique was self-standardized by trial and error method in the LG domestic Microwave (Model no. MS-285SD). The microwave was operated at the maximum output power of 40% (approximately 360 W) with rotating tray and ring removed. The cut piece from a fixed tissue sample was placed in a plastic tissue cassette and water washed in running tap water so that tissue was free of formalin. The tissue was irradiated in 200ml of 100% methanol and 200ml of 100% isopropyl alcohol for dehydration at cold spot for 2 cycles of 10 minutes each respectively in the microwave. After dehydration tissue was impregnated in 200ml of molten paraffin wax for 2 cycles at cold spot of 10 min each and was embedded in paraffin wax. Conventional Tissue Processing The cut piece from a fixed tissue sample was placed in a metal tissue cassette and water washed in running tap water so that tissue should be free of formalin. The tissue was dehydrated in 70% alcohol (one change), 90% alcohol (one change) and 100% alcohol three changes of 1 h each respectively. After dehydration tissue was cleared in two changes of xylene of 1 h each. Finally, tissue was impregnated in 2 changes of molten paraffin wax for 1 h each and was embedded. Rapid Manual Tissue Processing The cut piece from a fixed tissue sample was water washed in running tap water so that tissue should be free of formalin, after that tissue was wrapped in filter paper and dehydrated in 95% alcohol, 100% alcohol for 20 min on a stir plate. The dehydrated tissue was cleared in xylene for 20 min on a stir plate 20 min on a stir plate. Tissue was impregnated in 2 changes of molten paraffin wax of 1h each and was then embedded. The microwave processed tissue were stained as given in table no. ____. Conventional and rapid manual processed tissues were stained as given in table no.___________. Statistical Analysis The values obtained from different observers after assessment of sections processed by the three techniques were subjected to statistical analysis by Kruskal Wallis Test. One way ANOVA (Analysis of variance) was used for comparing mean shrinkage in tissues processed by the three histoprocessing methods. The P value Results All observers were assumed to be reliable as the Cronbach’s reliability test was statistically significant. Complete concordance was found amongst all pathologists in most of the cases. Hence, observer 1 was randomly selected for further analysis. The histopathological evaluation of the epithelium, fibrous tissues and glandular tissue revealed that the nuclear cytoplasmic contrast was good and cellular outline was distinct in tissues processed by microwave assisted technique followed by conventional processing and rapid manual processing techniques. The stroma was good with distinct cellular outline. The secretory products can be easily appreciated and the RBCs, inflammatory cells were intact. The results were statistically non-significant as observed by Kruskal Wallis test. The colour intensity of the tissues graded by four observers revealed that the microwave sections were crisper and there was a good contrast between the hematoxyphilic and eosinophillic areas. Though some slides were not visualised up to the mark, all the three histoprocessing methods were comparable to each other. One way Analysis of Variance (ANOVA) revealed a significant value on comparison of the tissue shrinkage processed by the three techniques. The dimensions of the tissues were recorded before dehydration and paraffin embedding. The mean percentage of shrinkage in rapid manual technique was significantly higher as compared to the other two techniques whereas statistically non-significant value was obtained on comparing conventional and microwave method of tissue processing. Discussion Microwaves are electromagnetic waves(Microwave Processing Techniques for Microscopy) which causes oscillation and excitation of polar molecules which are usually dipolar molecules of proteins in tissues. The heat produced enhances the rate of diffusion of fluids to permeate into the tissues. The rise in temperature decreases the viscosity of processing fluids that facilitates diffusion. Therefore it is theoretically possible to fasten the tissue fixation and processing. This has resulted in a substantial reduction in the basic steps of histoprocessing, thereby reducing turnaround time and providing same day diagnosis. The applications of microwaves are extensive which includes tissue fixation, stabilization of large specimens, tissue processing for light and electron microscopy, histochemical and immunohistochemical staining. Microwave tissue processing technique was introduced by Boon and Kok in 1985 (Leong 2004) but the potential application of microwave energy was first recognized by Mayers in 1970 who successfully fixed tissue with a microwave generator (Kok, Visser Boon 1988). Boon et al. (1986) reported that it was possible to produce significant acceleration of tissue processing by using microwave radiation. Visinoni et al. (1998) first described the tissue processor that completed the processing in 30-120 min, thus reducing the processing time from 24 h to just 1-2 h providing early reporting and easy patient management. Thus, the aim of the present study was to compare the cytoplasmic and nuclear details as well as staining characteristics of tissue sections processed by conventional, rapid manual and microwave techniques. The noxious chemicals used in conventional tissue processing were replaced in microwave tissue processing. In the microwave processing in contrast to conventional tissue processing, isopropyl alcohol was replaced by methanol as dehydrating agent and xylene by isopropyl alcohol as intermediate agent. Molten paraffin wax remained the impregnating and embedding medium for both the techniques. The reagent selection was in consonance with Babu, Malathi and Mangesh (2011) who also used methanol, isopropyl alcohol and molten paraffin wax for microwave tissue processing. Microwave radiation produced when enter the chamber it is reflected by the chamber walls until these gets absorbed by the material placed inside the chamber (Wong 2011). However, the spreading is not even throughout the chamber leading to formation of hot and cold spot zones. (Kok, Boon Smid 1993, Thostenson Chow 1999, Rutgers 2013). Hence, hot cold spots should be detected to achieve consistent results. Various authors have described methods for detection of hot and cold spots. Microwave processing was self-standardized by trial and error method in which the hot and cold spots were detected by using a damp thermal paper Kok, Boon and Smid (1993). All the procedures in the microwave were carried out in the cold spot zones as suggested by Sharp and Paperiello (1971), Benard (1974), Rangell and Keller (2000) in their respective studies. Microwavable plastic tissue cassettes were used for microwave tissue processing which are cheap and reusable as metallic utensils are contraindicated in the microwave because the electric fields of the waves produced by microwave magnetron are completely reflected at the same frequency by metals which can lead to sparking. (Vollmer 2004). In the present study, the staining protocol for microwave was followed as given by Babu, Malathi and Mangesh (2011) which included the stains used to be accelerated in the microwave. Kayser and Bubenzer (1990) used domestic microwave oven for acceleration of the various stains which also included HE stain. Valle (1986), Moorlag, Boon and Kok (1987) and Mathai et al. (2008) modified various special stain protocols for microwave and concluded that microwave did not produce any deleterious effects on staining. In our study, the three pieces of tissue processed by three techniques sectioned by a soft tissue microtome and stained as per their respective protocols were evaluated. We adopted the criterion for evaluation of tissue sections given by Kango and Deshmukh (2011). The overall quality of the tissue sections processed by microwave and manual methods was comparable. The microwave processed sections had same or similar cytoplasmic and nuclear details with good erythrocyte integrity and lymphocytic appearance than the manual methods. Similar results are given by Mathai et al. (2008), Morales et al. (2002), Bhuvanamha et al. (2013), Panja et al. (2007), Boon et al. (1986), Kok et al. (1988). We also observed that the stroma in some cases was slightly more condensed focally in microwave processed tissue sections which is similar to the findings reported by Boon, Kok and Ouwerkerk-Noordam (1986) This lead to the erroneous categorization of these cases as indistinct in studies by Kango and Deshmukh (2011). Since our criterion was adopted from the above mentioned study we also placed focal condensation of stroma as indistinct. In contrast Kok, Visser and Boon (1988) refuted the importance of focal condensation of stroma in diagnostic pathology. The colour intensity of the tissues graded by four observers revealed that the microwave sections were crisper and there was a good contrast between the hematoxyphilic and eosinophillic areas. The microwave processed tissues showed an increased reaction to HE. The sections stained were slightly more eosinophilic as compared to the manual techniques. Similar findings are reported by Hopwood et al. (1984), Boon et al. (1986), Chaudhari, Chattopadhyay and Dutta (2000), Leong and Price (2004), Panja et al. (2007), Mathai et al. (2008), Babu, Malathi and Mangesh (2011). Hopwood et al. (1984) suggested that this eosinophilia could be easily corrected by altering the stain composition or staining time in eosin. In contrast Leong Price (2004) observed that eosinophilia of the cytoplasm was advantageous as it produced good nuclear cytoplasmic contrast and enhancement of the cellular features. The dysplastic features i.e. hyperchromatism, pleomorphism of tumor cells and mitotic figures were easily appreciable in the microwave processed tissue sections of malignancy. There was also an easy appreciation of the giant cells in the tissues of Central Giant cell Granuloma and tubercular lymphadenitis processed by microwave processing technique. Rapid processing of histopathologic material is becoming increasingly desirable for intraoperative consultations and timely diagnosis. We found positive impact on turnaround time in microwave method as the time taken for block preparation from fixed tissue was 1h as compared to conventional method (9h) and rapid manual method (3h). In some cases, proper diagnosis could not be reached as the size of the tissue was small and the sample was not representative of the site. Similar difficulties were also encountered by Suri et al. (2006), Kango and Deshmukh (2011) in their respective studies. As assessed in our study, the effects of the three methods of histoprocessing on cytoplasmic and nuclear details of epithelial, fibrous and glandular tissue showed no statistically significant variation. The microwave technique was comparable or slightly better than the manual methods. Conclusion The applications and versatility of microwave processing methods are unattainable with conventional procedures. The method reported herein reproducibly yields similar histologic quality to that provided by conventional processing. It has many advantages including feasibility, safety and elimination of noxious chemicals that might be used for improvement in the practice of the histopathology laboratory, permitting the preparation of diagnostic material within a day. Domestic microwaves are easily available and cost effective but have certain notable disadvantages like uneven heating and inability to record and maintain temperature within the chamber. Further exploration in the field is required for development of cost effective microwave histoprocessors for histopathology which provide similar histologic material for rapid diagnosis and reporting.
